The env gene encoding the envelope protein type 1 of AIDS virus(HIV) and gp120 gene of Chinese strain of HIV 1 were inserted to the downstream of hybrid promotor of carrier(containing 16 P7.5 promotors of fowlpox virus mutant,late promotor of inclusion body type A of cowpox virus and LacZ labeled gene) constructed using the TK gene at unnecessary regions of fowlpox virus as flank, then 2 strains of recombinant chicken virus vUTA 2f 16LE and vUTA 2f 16LG were obtained by homologous recombination with wild fowlpox virus and selection of blue blot using X gal as substrate.Dot ELISA,IFA,SDS PAGE and Western blot demonstrated that both the viruses could express gp160 and gp120 with the molecular weights of 160 000 and 120 000, respectively. The immunization experiment in mice proved that the A vlue of antibody to gp120 in sera and the ratio of CD4 to CD8 in spleen were significantly higher than those in control group. It showed good reactogenicity and immunogenicity of expressed env and gp120 proteins.

The growth and metabolism of hybridoma cell in the scerum free culture was studied in 10L bioreactor.The results showed that the specific growth rate of cells reached maximum at the begining of logarithmic growth phase,then decreased gradually.Similar profiles were observed in the specific rates of oxygen consumption and ATP production.The specific rate of cell growth was linearly correlated with that of ATP production.It indicated the close relationship between the energy metabolism and cell growth.The authors considered that oxyg en uptake rate(OUR) could reflect cell metabolic activity timely and sensitively,and predict the change of viable cell density.It is concluded that OUR can be used for controlling feeding rate.

The influence of CH50,a recombinant polypeptide of human fibronectin,on the function of macrophages of mice during chemotherapy was observed.The result showed that continuously intraperitoneal injection with CH50 could prevent the reduction of the amount of peripheral blood monocytes and peritoneal cells caused by chemotherapeutic agent,enhance metabolic activity,cytotoxicity,the secretion of cytokines,and the proliferation of splenic lymphocyte.It indicated that CH50 could improve the curative effect of chemotherapeutic agent.

The normal mice and the mice with glucose hyperglycemia,as well as the rats with alloxan hyperglycemia were subcutaneously injected with recombinant human insulin.The biological value of the insulin was 28.2 U/mg.The dosages used for mice were 0.5,1.0 and 2.0 U/kg,and those for rats were 0.5 and 1.0 U/kg.The result showed that the blood sugar contents of all the animals were significantly decreased.The LD 50 of the insulin to male and famale mice were 21.69±2.46 and 47.56±6.10 U/kg respectively.The long term toxicity test in dogs showed no any toxicity of the insulin.\;

After the whole cells of helicobacter pylori was broken by ultrasonic wave,the urease in supernatant was purified by DEAE Sephe rose Fast Flow and Phenyl Sepherose CL 4B chromatographies.The reco very rate of urease was 30%,and the purification fold fo it reached 11.SDS PAGE proved that the purified urease consisted of 2 subunints with relative molecular weights of 60 000 and 30 000 respectively,and Western blot showed good antigenicity of it.The specific activity of the urease at 45℃ pH8.0 was about 265 U/mg.Three lots of urease wee purified,the result showed that the process was stable and laid a foundation of basic research and application of urease.\;

A novel tumor necrosis factor α(hTNF α)derivative named hTNFD was purified by heating treatment,salting out and ion exchange chromatography.The purity of the derivative after being purified reached above 96%,and the recovery rate and purification fold of it were 43.8% and 6.4, respectively.The detection result of L929 cell activity showed that,compared with hTNFα,the specific activity(1.01×10 10 U/mg) of hTNFD increased by 465 fold.The purification process was simple,stable and easy to be scaled up.

The F(ab') 2 fragments of murine monoclonal antibodies were prepared according to the clinincal quality standard. The fragments were purified from the McAb IgG 1 digested with pepsin by hydrophobic integration HPLC using phenyl separose high performance column. The purity and recovery rate of the fragments were above 98% and more than 50% respectively. The whole technological process was under the condition of sterility and free pyrogen. The product of F(ab') 2 fragment was stable at 4℃ for one year. The product reached the clinical quality standard published by National Institute for the Control of Pharmaceutical and Biological Products and was suitable for large scale production.

An ELISA kit was prepared with the antibody to NGF obtained by immunizing rabbits with the 2.5 sNGF purified from murine submaxillary glands. The sensitivity of the kit reached 1 ng/ml, and, at a range of 0.84 to 6.7 ng/ml, A value(absorbance) showed a good linear correlation with the contents of 2.5 sNGF(r>0.99). No non specific reaction was observed among the kit and the plasma fo human, mice and rats. The recovery rate of NGF from rat plasma was between 91% and 107%, and the CV of which was less than 10%(n=4). The NGF reference was assigned by the kit for 6 times, the result showed that content of it was 13.4 ±1.13 ng/dose, and the CVs of inter and intra assays were 8.43% (n=6) and 6% (n=4, 3 times) respetively. It proved that the kit could be used for the quantitative determination and pharmacokinetical study of murine 2.5 sNGF.

Inactivated hepatitis A(HA) vaccine was prepared by culturing the strain TZ84 of HA virus in 2BS cells, and the immunogencity of it was studied in common marmosets and Macaca mulattas. The result showed that good antibody respones were induced in the 2 kinds of monkeys. The vaccine could also protect the monkeys from the challenge with strong virus strain(Luxing). It indicated that the inactivated HA vaccine had good immunological effect.\;

The sialic acid contents in varous lots of the semi-finished recombinant Human erythropoietin(rhEPO) were detected by resorcinol color assay,then the relationship between the detection result and in vitro or in vivo biological activities of rhEPO was studied.The result showed the positive correlation between the sialic acid content and the in vivo biological activity of rhEPO. However, no influence of the content on the in vitro biologiccal activity of rhEPO was observed.

The specific CTL cell mediated immune responses induced by live attenuated and inactivated Japanese encephalitis(JE) vaccine in mice were compared. The result showed that the specific CTL activity induced by live JE vaccine was 79.2%, however, that induced by inactivated JE vaccine was only 29%. The neutralizing antibldy titer of mice immunized with live and inactivated JE vaccine were 1:5 and 1:20 respectively , and the immune potency of the 2 kinds of vaccine were 3.6×10 -6 and 4.0×10 -4 -4.2×10 -4 ID 50 /ml , respectively. It indicated that the cell immunity induced by live JE vaccine played an important role in protective potency.

Eighteen patients with acute aplastic anemia(AAA) were divided into 2 groups. The 8 patients in treatment group were intravenously injected with rhGM-CSF for 2w at dosages of 5-15 μg/kg·d on the basis of routine treatment.However, the 10 patients in control group only received routine treatment. The result showed that, compared with control group, the amount of peripheral blood leukocytes of treatment group increased by 5-30 folds. rhGM-CSF could abate the clinical symptoms of AAA and reduce the opportunity of infection. So it is an effective assisted drug for AAA.

In order to study on the reactogenicity and immunogenicity of simultaneous administration of MMR and hepatitis B vaccine, the children aged 8-13 years were simultaneously or separately immunized with the 2 kinds of vaccines. The result showed that both the adverse reactions caused by simltaneous and separate immunization were mild, and the response levels to hepatitis B,mumps and measles were similar. So authors considered the MMR and hepatitis B vaccines could been administered simultaneously.
