In order to study on the potential effect of p53 tumor suppressor gene, a novel method for gene transfection was developed.The plasmid pCEP4-p53 containing the cDNA of wild type p53 gene was combined with cation liposome and transferrin,then transfected to LTEP-P cells. Cell apoptosis was proved by microscopy,acridine orange (AO) staining,agarose gel electrophoresis and nick end labeling test.The result showed that tumor cells were successfully transfected with the DNA-liposome-transferrin triple complex in vitro, and the apoptosis of tumor cells were induced by wild type p53 gene.

A novel double induced expression system containing plasmids pJGW1 hGCSF and pGP1 2 was constructed on the basis of study on the expression efficiency of recombinant hG CSF gene and the genetic stability of enginerring E.coli.After the recombinant bacterial strain,pGP1 2 pJGW1 hGCSF/DH5α,was induced at 42℃,27% of hG CSF was expressed.The recombinant bacterial strain had been undergone 140 passages,the result showed the good genetic stability of it.

The NS5 gene fragment of HCV was amplified from anti HCV and HCV RNA positive sera by RT PCR.After the gene fragment was inserted into expression vector pET 28a and transformed to E.coli BL21(DE3),NS5 protein was successfully expressed and purified by immobilized metal affinity chromography(IMAC).Western blot and ELISA proved that the NS5 protein was specifically reacted with the sera of patients with hepatitis C and might be applied for the diagnosis of HCV infection.

The strain L A 1/P9 of hepatitis A virus was cultured and passaged in human embryo lung diploid cells(2BS strain) at 35℃, then cloned and purified. In this way, a new candidate strain of live attenuated hepatitis A vaccine was selected out. The candidate strain passed foreign factor examination. The result of rhesus test showed good immunogenicity of the candidate in comparison with the current strain, and no significant difference was observed between the safeties of them. The titer of HA vaccine prepared with the candidate increased by more than 1.0 Log compared with that prepared with the current strain, however, the virus proliferating cycle of it decreased by more than 10days.

A method for preparing experimental ISCOMs vaccine with human cytomegalovirus(HCMV)enveloped protein and stimulating cellular immune response in mice was reported in this study.No significant increase of Tc and NK cell activities were observed in the mice vaccinated with 1 dose of ISCOMs,but the rate of L3T4 and L3T4/LyT2 were significantly increased.However,after the 2nd dose was given,the Tc and NK cell activities of the mice were significantly increased.It indicated the potential preventive effect of the ISCOMs on primary HCMV infection.

In order to study on the effect of low molecular weight substance extracted from neonatal calf brain on neurite formation, the substance was added in the 2 serum free media in which chick embryo dorsal root ganglion and neonatal rat brain cells were cultured respectively. The result showed that neurite began to grow around chick embryo dorsal root ganglion 48 h after the substance was added in, and became long and dense until cross linked as time goes on. The average number and length of neurites of neonatal rat brain cells were also significantly increased 24 h after the substance was added in. It proved that the low molecular weight substance could promote neurite formation. Five peaks were observed on the HPLC profile of the substance at the wavelength of 280 nm.The effect of each fraction should be further studied.

A strain of recombinant E.coli expressing rhGM CSF fusion protein containing thrombin restriction site was selected out on the basis of comparing the characteristics of 4 strains.The molecular weight of the expressed product was 26000,and the N and T terminals of it were MS2(11000)and rhGM CSF(15000), respectively.The goal protein was released easily after the expressed product was digested with thrombin and purified by anion exchange or hydrophobic and/or gel filtration chromatographies.The result showed that the purity of it reached 10 7 IU/mg.

The parathyroid hormone related protein(PTHrP) with 6 histidine residues at its N terminal was inserted into pBV222 vector and expressed in E.coli, then purified by immobilized metal affinity chromography(IMAC).The recombinant PTHrP showed good biological activity after one step purification, and the purity of it reached above 90%. McAb was prepared with the purified PTHrP. The application of IMAC made the purification process of PTHrP simple and laid a foundation of study on the biological acitivity and clinical effect of the protein.

More than 25 000 serum samples were collected from 14 provinces, autonomous region and municipalities directly under the central authorities of China. Of the samples, 264 were retested by 14 kinds of anti-HCV EIA kits made in China and 2 kinds of the 3rd generation of the kits made in America, then confirmed by RIBA HCV 3.0 and PCR reagents. After 2 times of collabortive assignments by 16 Chinese and 2 American laboratories, 40 positive and 40 negative samples were selected out and used for preparing the 3rd generation of national reference for anti-HCV diagnostic kits. The reference has been evaluated by the expert commitee and approved by the Ministry of Public Health. Clinical trial proved that the reference promoted the improvement of the qualities of national kits and reached the international level.

The cytotoxic effect of combined administration of tea pigment and recombinant human tumor necrosin factor(rTNF) on primary liver cancer cell was detected by MTT colorimetric assay. When tea pigment or rTNF was administered alone, no significant cytotoxic or inhibition effects of tea pigment on liver cancer cell H7402 were observed,and the maximal inhibition rate of rTNF on the cell was 35.1%.However, the maximal inhibition rate of combined administration of the 2 drugs reached 47.2%. The result showed the the cytotoxic effect of rTNF on H7402 cell could be improved by tea pigment.

The adverse reactions was observed in 140 elderly person (aged 55 70 years) 6-8,24 and 72h after being intramuscularly injected with 23-valent(575μg/0.5ml)penumococcal polysaccharidevaccine.The systemic reaction was mild. Local red and sswelling at different degrees were observed in 28(20.00%) and 38 cases(27.14%) within 6-8 and 24h respectively, while local pain were observed in 58(41.42%) and 41(29.28%) cases respectively. However, all the reactions were abated 48h and disappeared 72h after immunization. Blood samples were collected from 100 of the 140 person before and 1 month after immunization and detected for the titers of 23 kinds of antibodies by improved ELISA.The result showed that the GMT of total antibody of them increased by 2.4 folds. Significant differences were observed in the titers of all the antibodies except for type 3 before and after immunization. It showed good immunogenicity of the vaccine.
