A recombinant plasmid(pGEX p24) was constructed by inserting the capsid protein gene P24 amplified from HIV 1 gag genome by PCR into a expression vector pGEX p24 and transfered to E.coli. After being purified by Glutathione Sepharose 4B affinity chromatography, the expressed P24 was detected by ELISA and Western blot and showed good antigenic specificity and immunoreactivity.

After the total RNA was extracted from fetal liver,the whole length gene(1 080 bp) of human thrombopoietin(TPO) was obtained by reverse transcription and PCR. The TPO gene was digested with Nde Ⅰ and Hind Ⅲ,then inserted into pRSET A vector and transformed to E.coli HMS174. The 2 positive clones obtained were identical in DNA sequences. On the basis of transfection of E.coli JM109(DE3) with positive plasmid, the expression of TPO was induced by 1 mmol/L IPTG. SDS PAGE showed that the molecular weight of the expressed TPO was about 40 000.

Recombinant baculoviruse was constructed by inserting a full length cDNA open reading frame( ORF) coding HAV capsid protein into the end of polyhedrin promoter and expressed in SF9 cell.The result proved that the recombinant baculoviruse contained a full length HAV ORF,and the expressed protein showed good antigenicity and immunogenicity.HAV particle like spherical structure was observed in the SF9 cells infected with the recombinant baculoviruse under electron microscope.The buoyant density of the recombinant protein detected by cesium chloride gradient centrifugation was similar to that of HAV capsid.

After secretory human growth hormone(rhGH) engineering bacterial strain W3100 was cultured in fermenter, cells were lysated by osmotic shock method, then rhGH was purified by ion exchange, hydrophobic and molecular size charomotagraphys. The purity and specific activity of the purified rhGH reached above 97% and 2.5 IU/mg respectively.

After the total RNA was extracted out from the aortic smooth muscle of rabbits with arteriosclerosis, the gene coding platelet derived growth factor A was amplified by RT PCR. On the basis of this, clone(SK/PDGF) and expression(PKK/PDGF) vectors were constructed, and PDGF A was successfully expressed.

Yolk antibody(IgY) was purified from the eggs yolk laid by the hens immunized with the dental caries vaccine DCV strain(constructed by the authors) or glucosyltransferase(GTF) antigen(extracted from DCV strain) by water extraction, salting out with ammonium sulfate and DEAE Sepharose column chromatography. The antibodies to GTF were detected out by RIA method both in the serum and eggs of the hens. SDS PAGE showed that the molecular weight of the purified IgY was about 220 000, and the purity of it reached 97.9%. After being treated with 2 mercaptoethanol, the IgY was dissociated into a heavy(76 000) and a light(28 000) chains. Western blot showed the good immunological specificity of the IgY.

The purity of McAb1A 1 to acute nonlymphocytic M 5 leukemia reached 99.9% after being purified by ion exchange and gel filtration chromatographys. Three kinds of immunoconjugates, McAb1A 1 DNR, McAb1A 1 CADNR and McAb1A 1 PAD DNR, were prepared by direct coupling, cis aconity/linkage and indirect coupling respectively. The molar ratios of the 3 immunoconjugates were 1∶52, 1∶27 and 1∶437 respectively, and 84.6%,50% and 91.6% of drug activities of them were retained respectively. Though the antibody activitiy of McAb1A 1 PAD DNR was significantly decreased, no significant decrease was observed in those of McAb1A 1 CADNR and McAb1A 1 PAD DNR. McAb1A 1 PAD DNR was the most effective in killing U 937 target cells. The authors found that Lowry method could not be used for determining the antibody protein contents in the conjugates containing DNR, and live cell ELISA could not be used for determining the antibody acitivities in them.

The influence of culture temperature on the growth density of recombinant bacterial strain(pCG 1/rhG CSF/DH5α) and the expression of rhG CSF were studied. After the temperature during culture was changed, both the growth density of pCG 1/rhG CSF/DH5α and the yield of expressed product were improved, and the culture time was shortened. Three lots of pCG 1/rhG CSF/DH5α were cultured in NBS MPP 40 fermenter for 15 h, the result showed that the density(A 600 ) and dry weight of the bacterial strain were 25.0 and 23 g/L respectively, and 26.93% of rhG CSF was expressed.

The rats with hyphemia caused by chronic renal failure were divided into 3 groups and subcutaneously injected with recombinant human erythropoietin(rHuEPO) for 14 days at the doses of 450, 150 and 50 IU/kg respectively, using EPOGEN(Amgen,USA) and normal saline as control. The rHuEPO showed curative effect on the hyphemia caused by chronic renal failure. However, it showed no effect on chronic renal failure.

Living cells can be stained by crystalline violet. Depending on this principle, a colorimetric method for determining the potency of IFN was developed. The potency was calula ted by parallel line assay and compared with that determined by routine method. The result showed a good correlation between them (r=0 9387) . The cell pathological change expressed as A value was more accurate than that expressed as score. Therefore, the new method is more accurate and reliable and can be used for the determination of IFN.

A new method for screening and evaluation of immunomodulator, CD + 2 cell proliferation test, was established. After the CD 2 antigens on the surface of peripheral blood mononuclear cells(PBMC) were digested with trypsin, the PBMCs were incubated at 37℃ together with immunomodulator for a appropriate time, then the CD + 2 cell proliferation rate was detected by biotin streptavidin immunocytochemistry method and used for evaluation the activity of immunomodulator. The activities of several kinds of known immunopotentiators and immunosuppressants were detected by the method, and the optimal concentractions of trypsin and the optimal time for digestion and incubating were studied. The method was simple and showed good reproducibility. It was also easy to be standardized.

A reference for determining the activity of recombinant staphylokinase was prepared according to the requirements for standard.All the detection indexes of the reference met the requirements for standard.The result of 15 times of assignment test showed that the activity of the reference was (1010±191) U/does with a variation coefficent of 19%.The reference also showed good stability.
