Protein P_(110) encoded by oncogene of MGC803 cells of human gastric carcinoma cell line was purified as an immunogen to immunize the hens. IgY, a specific antibody to human gastric carcinoma, was obtained from the yolk of eggs laid by the immunized hens. The rate of immunohisto chemical reaction between the antibody and human gastric carcinotissue was 80%, that is obviously higher than the reaction rate with other carcinotissue and normal gastric tissue. The IgY was cross linked with the chain A of ricin, termed as IgY-Ricin A, which was used to attack human gastric carci noma cells and other carcinoma cells in culture. The results showed that the TCID of human gastric car cinoma ceils was 0. 01mg/ml, obviously lower than that of other carcinoma cells. It suggested that IgY was a better polyclonal antibody directed against carcinoma.

1C_6 McAb_2 is a syngenic murine monoelonal anti-body to the idiotype of 7A_4, monoclon al antibody (McAb_1). 7A_4 McAb_1, were extracted and purified from murine ascites by (NH_4)_2 SO_4 pre cipitation and 1C_6 McAb_2-Sepharose 4B affinity chromatography. Purified protein was reduced by 2 -mercaptoethanol was identified by SDS-PAGE. The result showed two bands which were heavy chain and light chain of IgG respectively. It was observed that a specific precipitation line with sera of rabbit against mice IgG and a single precipitation are with sera of rabbit against mice sera when double diffusion and immunoelectrophoresis were performed. The PHA titre of purified McAb reached 125 000/mg. The McAb purified by this method resulted in high yield, good purity and strong specificity.

The high density suspended culture of Hela cells was performed by increase the amount of changing medium in Celligen Bioreactor. The result showed that the density of cells reached 1.1×10~7/ml, and more than 10~(10) viable cells could be acquired each week.

An ELISA kit using polyvalent antign (A59, MHV1 and Sh-79) screened out from 6 strains of MHV was prepared. 104 serum samples of SPF and clean level mice were detected for MHV antibody by the kit and the result showed that x, SE and positive cut-off value were 0. 037, 0. 024 and 0. 207, respectively. The specifcity of the ELISA kit was detected by electron mcroscope, blocking inhibition test and IFA test and proved to be good. The sensitvity of detection of MHV antibody by the ELISA kit was higher than that of by monovalent antigen. The positive rate of antibodies of ordinary mice was 72% (161/223), and that of mice of class I and Ⅲ was 1.7%(2/ 117).

Freeze dried human leucocyte γ-IFN was studied on its heat stability at four different temperatures. The result showed that the residual activity was more than 50% after the preparation had been heated at 66℃ for 11 days, or 56℃ for 35 days or 37℃ for 5 months. The losses of activi ty were less than 10% and 15% after the preparation had been stored at 5℃ for one year and two years, respectively. The calculation results obtained from the formula associated with accelerated degradation tallyed with that obtained from the tests above. Thus, the heat stability of the preparation was good.

During the course of detoxification of purified diphtheria toxin by using formalin alone, the average of loss rates of Lf/ml and Lf/mg PN were 14.81% and 13%, respectively. However, when dextoxified by formaln and lysine, no Lf/ml was lost. On the contrary, Lf/mg PN was increased at a average rate of 6.74%, and the average of purity of the product reached 2216 Lf/ mg PN. After the product had been ultrafiltrated by hollow fiber column with an cut-off molecular weight 30 000, its purity was increased at a rate of 15. 21%, and reached 2540 Lf/mgPN in average. The results detected by HPLC showed that the rate of specific component of the vaccine rose from 78. 26% to 91.36%, and the rate of nonspecific component descended from 21.74% to 8. 64%, and the molecular weight of specific component was about 64 500.

A method of determining the content of the octanoic acid in human plasma albumin by gas chromatograph (GC) was repoted. The samples added in heptanoic acid as internal standard were previously treated by perchloric acid, extrated by chloroform and concentraed, then detected by GC. A graph was prepared by plotting the ratio of peak area of octanoic acid as abscissa and ordinate, respectively, and a regression equation was obtained from the graph. The content of actanoic acid could be calculated from the ratio of peak area of octanoic acid to heptanoic acid in samples by the regression. The recovery rate in this assay was 96.88±1.19%, and the coefficient of variation in 17 batches of products detected by this method for 4 times averaged 2.72% (1.21~4.62%). The method was relatively simple and convenient to handle and could be used to control the content of octanoid acid in human plasma albumin.

The samples of rIL-2 were first separated by vertical SDS-PAGE according to their molecular weight, then transferred to nitrocellulose membrane by using electron transfer system. After rest of the sites were blocked by calf serum or bovin serum albumin, murine anti-human IL-2 McAb (DMS-1) were added, and stand for 1.5h at room temperature. The membrane was washed and rabbit anti-routine IgG-HRP was added, then stained by DAB. The specificity and purity of the samples were identified by the position and amount of bands respectively. The result showed that there was only a single band on the membrane with a molecular weight of 15 000, and no polymer of protein was found.

Two inactivated vaccines, i. e. CP89-1(China)and COPARVAX4096 vaccines (British) were examined by sterility test, toxicity test, pyrogenicity test, activated spleen test and tu- mor inhibition test. The results showed that the quality of both vaccines could meet the Chinese Re quirements for the Biological Products. The spleen index and the rate of protection to mice in tumor in- hibition test of CP89-1 were higher than that of COPARVAX4096 vaccine.

Anti-HCG monoclonal antibodies were prepared by intrasplenic direct immunization. The titer of the antiserum detected by FLISA was above 1:150, and the titer of the monoclonal antibody detected by the same method was above 1:6000. This method were simple and convenient to handle, and the dose of antigen and time were both saved compared with common immunization method. It had no difference in titers and specifities of the McAbs obtained between direct and common immunization method.

58 645 children aged 1~7 were selected for an epidemiologic study on the efficacy of Japanese Encephalitis (JE) live vaccine in 1991. Only 2 of the 29 639 vaccinated children suffered from JE, the morbidity was 6.75/100 000. In the unvaccinated group, 46 among 29 006 children contracted the disease and the morbidity was 158.59/100 000. The protective ratio and the protective rate of the live vaccine were 1:23. 49 and 95. 74%, respectively. It showed significant difference be tween the two groups (x~2=39. 49, p<0. 005). The result indicated that the protective efficacy of JE live vaccine was superior to that of JE live vaccine as reported in this country. The vaccine showed good effect on the severity and prognostic of JE. The live vaccine is also convenient and safe to adminstrate.

From 1983 to 1991, 17 524 newborns resided in Hangzhou City, Dalian City and Liaoning Provinces were vaccinated with 6 BCG trial products made from different strains: Brazil, Bei- jing, Japan, Changchun-Shanghai, Shanghai and Denmark and with 5 BCG final products manufac tured by 5 Institutes of Biological Products from Shanghai, Beijing, Changchun, Chengdu and Lanzhou. The immune responses were detected by skin test with PPD. In Dalian, the results showed that the 12-weeks-positive rates of BCG trial products made from Shanghai and Brazil strain were the highest, and that of BCG trial product made from Beijing strain was the lowest. The 5-years-positive rate of BCG trial products made from Shanghai strain was the highest, and that of BCG trial product made from Beijing strain was the lowest. In Hangzhou, the results also showed that the immune re sponse of BCG trial products made from Shanghai and Japan strains were the best, and that of BCG tri al product made from Beijing strain was the worst. In Hangzhou and Liaoning, the results showed that the 12-weeks-positive rate of BCG final product manufactured by Shanghai Institute of Biological Prod- ucts was the highest, and that of BCG final product manufactured by Lanzhou Institute of Biological Products was the lowest. The BCG final product from Shanghai Institute might lead to lymph node suppuration when the viable count of bacteria was high in some lots, but the proba- bility of occurrence was not more than 1.
