  • The Prophylactic and Therapeutic Effect of Human High Density Lipoprotein on Experimental Atherosclerosis in Rabbits

    We studied the effect of human high density lipoprotein on experimental athe-rosclerosis in rabbits.The results showed that the human high density lipoprotein can prevent the cholesterolfrom attaching to the intima of arteries,and diminishes the severity of fatty plaques for-mation.For prophylaxis,when human HDL and high dose cholesterol simultaneouslywere admi-nistered intravenously to rabbits,the fatty plaques presented in only 1.6% of the totalarea of arteries as compared to 15% in control animals.With the HDL administered subsequent to the formation of experimental fatty plaques,the histological studies revealed no pathological changes remained,whereas half of the ar-terial intima surface being packed with fatty plaques.

  • Antibody Responses of Inactivated HFRS Vaccine to Homologous and Heterologous Types of Virus

    The antibody responses of HFRS Z_(10) virus and its vaccines to homologous andheteroiogous types of viruses were studied.The results revealed that rabbits immunized withboth live virus and inactivated vaccine elicited neutralizing and HI antibodies to homo-logous viruses ranging from 4 to 8 fold higher than the antibodies to heterologous viruses.10 volunteers vaccinated with the Z_(10) inactivated vaccine showed higher NT and HI anti-bodies to the homologous virus and the seroconversion rates to the homologous virus was 90%(both NT and HI antibodies),but to the heteroiogous otis viruses only 20~30%.This results provided the evidence of antigenic difference between apodemus-type andrattus-type HFRS viruses.It also suggested the necessities of developing bivalent vaccinewith the two sero-types of viruses or selecting an antigenically broad virus strain as a can-didate vaccine virus strain.

  • Establishment and Identification of Monoclonal Antibody Hybridoma Cell Lines of Anti-HBe and Anti-HBc

    We successfully established 4 hybridoma cell lines of anti-HBe and 4hybridoma cell lines of anti-HBc,and the specificity and titer of the monoclonal anti-bodies secreted by the cell lines were not influenced after cultured for 3 months con-tinually.Specificity test showed that the monoclonal antibody has very high speciality of anti-HBe and anti-HBc.The class and subclass of Ig and chromasome of hybridoma cells werealso identified.

  • Study on Purified Japanese Encephalitis Virus Cell Culture Vaccine

    The original inactivated Japanese encephalitis vaccine was clarified and con-centrated to 10~20 times by hollow fiber ultra-filtration.Pelleting by the ultracentrifuga-tion method was chosen for further purification.The potency of the purified vaccine wassignificantly improved,and reached the level of the Japanese BIKEN mouse brain derivedvaccine.A preliminary survey showed the HI antibody response was 100% 4~x rise (GMT258.9),with very significant difference comparing with the original vaccine.

  • Irreversible Toxoiding of the Purified Diphtheria Toxin

    The purified diphtheria toxin was converted irreversibly into purified toxoidwhen it had been incubated for a adequate time at 37℃ in the presence of 0.0125 mol ly-sine and 0.5% formalin.The purified diphtheria toxin detoxified with formalin alone showed some reversion oftoxicity when the free formaldehyde in it was removed,and thus the irreversible toxoidingcould not be achieved.In fact,the free formaldehyde has to be maintained to inhibit thereversion of toxicity of the preparation detoxified without lysine,but it always causes ex-cess detoxification on storage and results in damaging the potency of this preparation.The antigenicity,stability of the potency,the adsorption rate on to Al (OH)_3,espe-cially on to AlPO_4,and the rate of lose in Lf/ml in the course of detoxification of lysine-preparation were all superior to the preparation detoxified without lysine.The ~3H-lysine incorporation test showed that the lysine has been conjugated into theprotein of the purified diphtheria toxoid in the course of detoxification.

  • Preliminary Report on Preparation of HuLeTF Using the Leucocyte Precipitate after Preparation of HuleIFN

    Since the collection of human leucocyte is rather difficult,production of Hu-LeTF is limited.4 small hatches of HuLeTF have been prepared with the leucocyte pre-cipitate after preparation of HuLeIFN Each lot was divided into two portions for acid treat-ment and non-acid treatment.By biological and biochemical examination,the ratio valuesof E260 nm/E280nm for all the samples except one attain at and are overl.8.Other qualitycontrol results could all reach and conform to the provisional requirements for productionand testing of HuLeTF,as well as regulations referred.The mean recovery was 1 ml TF/6.7×10~8 cells.It is about 36% of recovery of normal leucocytes.The mean concentration and contents of polypeptide and nucleotide of the samples withacid treatment were 50%,32.9% and 43% lower than that of non-acid treatment.The ratiosbetweenacid and non-acid treatment showed no signifficant difference.The infective particlesof NDV were effectively inactivated by means of acid treatment and intercepted by theultrafi ltration system.The preliminary results showed that it would be feasible to produce HuLeTF using the leucocyteprecipitate.

  • The Reaction and Efficacy of a New Oral Cholera Vaccine in Volunteers

    A new type of cholera vaccine which contains genetically engineered B sub-unit of cholera toxin (BS) and killed whole vibrio cells (WC) has been administered orallyto the volunteers to evaluate the side effects and immunogenicity.The new vaccine hasbeen proved to be safe and immunogenic in human.The vaccine exhibited two to fourfoldhigher rise in titer of vibriocidl antibody and fourfold higher rise in titer of antitoxin (aCT)than the control group.The efficacy of this new vaccine was comparable to the vaccinewhich composed of (BS) purified from nutural cholera toxin and WC.The results suggestedthat the new BS-WC oral vaccine could be a good candidate in cholera prophylaxis.

  • Comparison between HI and NT Antibody after Vaccination with Measles Vaccine

    The relationship between HI and NT antibody (Ab) after vaccination withmeasles vaccine has been investigated in a closed region without interference of wild mea-sles virus.The results indicated that the NT Ab could be detected in 82% and 43.8% sub-jects respectively when their HI Ab had converted continuously into negative (<1∶2) for≤5 years and 6~10 years.But for such a correlation,there was no difference between pri-mary and revaccination groups.This result showed that HI Ab titer <1∶2 is bv no meanswithout immunity,and the Ab would be solid after successful primary inoculation.Among the vaccinated subjects,3 years after infected with clinical or subclinical mea-sles,possessed higher titer than revaccinated subjects in both HI and NT Ab at the same in-terval.

  • Study on the Protective Effect of Dysentery Vaccine with a Model of Keratoconjuntiva in Rabbit

    The study on the protective effect of oral dysentery vaccines that made fromsd strain of flexneri 2a and flexneri 3a with rabbit keratoconjuntiva model.The resultsshowed that the complete protective rates were 28.5% and 19.1%;the partial protectiverates were 47.6% and 61.9%;the shortening rates of courseof disease were 55.0% and 47.7%respectively.These results compared to control with Ridit Analysis,u=3.95 to 9.48,P<0.001.Its overall protective rates were 80.5% and 76.5% respectively.The test showed thatthis model was suitable and cheaper and its experimental condition was easier undercontrol than primate.Futhermore it would not have any early infection.It was easier inobtaining and feeding and was more convenient for operation and observation than that ofguirea pig.Therefore,this model seems worthy for further study and popularization.

  • Report on the Survey of an Outbreak Epidemic of Adult Diphtheria

    An outbreak of diphtheria in adults occurred in a rural area of Wuxi citywhere the disease had been absent for 11 years.5 adult diphtheria cases and 2 carriers appeared in a household with 8 members fromOct.15 to 21 1988.After that 5 cases were reported in the epidemic village up to Dec.281988.Troat swabs of 41 villagers were taken and cultured for carrier examination and 3persons were found to beoositive in C.diphtheria (all toxigenic strains).329 people adjacentto the epidemic village were examined by Schick test before immediate immunization cam-paign and it was found that the positivity was 25.76% (34/132) in 25~39 year groupscompared to 2.54% (5/197) in 2~15 year groups (x~2=40.78,p<0.001).These facts suggested that revaccination for diphtheria toxoid in adults should be con-sidered.
