  • Variation among Varicella-Zoster Virus DNAs in Restriction Endonuclease Cleavage Patterns

    The DNAs of seven varicella-zoster virus(VZV)isolates were extraeted by disrup- tion of the purified viral nueleocapsids.The VZV DNAs were cleaved by two restriction end- onueleases.and analysed by agarose gel e’e trophores s.In the gel pattern of VZV DNAs cleaved with EcoRI,variations appeared among EcoRI-G,EcoRI-J and EcoRI-p fragments.The here- rogeneity in the gel pattern of VZV DNAs cleaved with BamHI was shown mainly in fragments with low molecular weight.The comprehensive compatison of the gel patterns of the tested VZV DNAs indicated that the seven isolates were non-homologous.

  • Application of DNA Molecular Hybridization Technique in Quality Control of HBV Plasma Vaccine

    By Using 32P—labeled whole genomic DNA of HBV ayw as a probe,the samples collected from partial C011ection of surose rate centrifugation during the preparation of HBV Plasma vace- ine and collected randomly samples from final bulk were analysed.The result showed by Mole- cular hybridization that all of the samples containing whethef long tubular form particles or short tubular form particles obseved under electromicroscope are negative,but the samples con- taining Danes’particles form 3 lots of the preparation including 6 collections are positive.This result indicates that the presence of long tubular form Particles evaluated by electromicroscope as samples non-passed needs to be furtller discussion.The sensitivity of the HBV DNA probe call reach 1pg.

  • The Discovery of a Large Plasmid in Rough Type Strain of Sh.sonnei

    A large plasmid was discovered in a rough type strain of Sh.sonnei named as S_7R.The presence of the large plasmid was reported only in S form of Sh.sonnei strains, which was considered relating to the I form antigen before. The plasmld is about 64 Md in size and 56 Md smaller than I form large plasmid(120Md)in wild type,which is thought to be the deletion mutant of the wild type plasmid and was iden- tified and confirmed by experiments in our 1aboratory.It was shown that the gene regions encoding 1virulence and I form antigen were deleted from the original plasmid by about 56Md in 1ength. The large plasid in the parent S_7 strain of the S_7R type is different from S_7R and the Wild strain.It is 10Md bigger than S_7R and 46Md smaller than the wild one.It encodes I form alltigen efficiently but shows no virulence.Therefore S_7 strain cohld be used as a donor to cons- trut I form antigen-containing dysentery vaccine safely. The discovery of the large ptasmid in S_7R would be benefit to the genetical study of I form large plasmid and I form antigen.

  • Serological Typing among 712 Clinical Isolates of Streptococcus Pneumoniae

    712 clinical isolates of S pneumoniae was serogrouped into 42 groups or Lypes by“quel- lung”reaction with lyping sera among which types 40,42,44 and 47 had not been found in Chlna before.of the 712 strains,type(group)5,6,1,19,23,2,14 and 3 were the most frequently encountered ones and these eight types(groups)totaled 453 strains(63.3%)and type 16A and 10C are found as new types. of the 180 isolates ftom CSF,137 strains(76.1%)of type(group)2,5,6,1,27,23,14,12 and 7 were the common estones;52 of 71 strains from blood(72.6%)the most common types(groups) were 1,5,6,14,19,7,8 and 35,183 of 253 strains(72.3%)isolated from otitis fluids.The most common types(groups)were 19,6,5,23,14,3,1 and 15.

  • Gene Sequencing of Recombinant Human IFN-α1 after High Density Fermentation

    In order to veelfy the Subtype of IFN-α-1 structural gene in engineeing strain BMH_(71)-18/ pBV857 and to monitor any change of the gene that might happen after felmentation,we isolate its structural gene form harvestied culture via high density fermentation and then sequence it by the Sanger dideoxynucleotide chain termination method and by the Ray method based on adding separately synthesised primers.In eomparisoh of this Sequence with the sequece of original strain and the sequence reported in 1iterature,we did not find any change of base pair.

  • Virological Survey of China Rhesus Monkeys

    A virological survey of 17 viruses and/or antibodies in rhesus monkeys were carried out This repors presents data based on 16,321 necropsies,including 6,715 virus isolations,8,932 neurohistologlcal tests,and 7,379 serologlogcal surveys.SFV,SV40 and their antibodles were. demonstrated most frequently,the next most frequent were antibodies to measles and vaccinia viruses.No B viras was found,but there were 12.89% of antibodies to HSV-1.This should be carefully evaluated.

  • Study of Human Tetanus Immune Globulin

    After 2-3 muscular iniections with adsorbed tetanus toxoid as basic immunjzation, the donors were further hyperimmunized with a big dose(36Lf/0.5ml)of thepurfied reranus toxoid.Accordinly,a plenty amount of immune plasma with high antibody titer was determined with ID techn que in vitro. From above pooling plasma we prepared three barches of TIG.The antibody titer of TIG was 260-330IU/ml which was concentrated 13-18 times compared witl poeling plasma.The stability of TIG was desirable as well. In first stage of clinical trial for 20 adults with 250IU/m1 TIG,all the persons have shown satisfactory startding to TIG without any Io al and all over rea tions.For second stage, the same dosage of TIG Was intected to 156 cases snffering from different open tratma.The resnlts has given significant evidence for prevention tetams with TIG.The level of antibodies in ser also indicated that TIG is better than TAT. Our results have demonstrated that TIG was effective,reliable and very safe for not only prevention but also therapy of tetanus disease.

  • Antibody Persistence after Immunization with HB Vaccine

    282 HBV seronegative children aged 8-11 were ineluded in the trial.151 children were vaccinated with 10μg of HBsAg in one dose on month 0,1,2.Other 131 children Were immunized on month 0,1,6.Former anti-HBs seroconvers ons rate at 3 and 36 months were 96.5% and 83.7%,respertively.This result showed that about 12.8% vaceine responders have been negative,while later no changed for 24 months,In anti-HBs level,ant-HBs peak for former was found at 6 months after intial vaccination,but at 7-12 months for later.Antiody level at 24 months was 3.5 times of former. Formet anti-HBs at 12 months decreased 4.3 times from that at 6 months.After 24 months ant-HBs 1evel decreased slowly.Ant-HBs value at 36 months was still maintained over 70.3 mIU.The results support that the persistence of anti-HBs after the Vaccination at least is over 3 years.

  • Lasting Time of Placenta Transfering Measles Antibody in Infants

    The author observed the lasting time of placenta transferng measles antibody in infants.The titre of placenta transfering antibody began to drop month by month from the fourth month after birth.In the eighth month,the antibody couldn’t be detected in 93.33% of the infants.The month on which the first immunization of live measles vaccine should be taken according to the results of study and the conditions of our country were discussed.
