• Cloning, Sequencing and Expression of Glycoprotein Gene of Chinese Street Strain(SBD07) of Rabies Virus

    After the good immunogenicity of Chinese street strain(SBD07) of rabies virus had been proved, the glycoprotein gene of the strain was cloned and sequenced by RT PCR. The result showed that nucleotide and protein homologies between the genes of SBD07 and vaccine strain(aG) were 85.8% and 88.5% respectively. The glycoprotein gene of SBD07 was successfully expressed using vaccinia virus as vector. The expressed product could induce high level of neutralizing antibodies to rabies virus in mice and protect them from lethal challenge of rabies virus CVS strain intramuscularly.

  • Gene Cloning, Expression and Immunology of Tetanus Toxin Fragment C

    The fragment C gene of tetanus toxin was amplified from plasmid DNA of Clostridium tetani by PCR. deteced by DNA sequencing, then cloned into vector pGEX 4T 2 to construct a recombinant plasmid pGEX TC。 SPS PAGE showed that a specific recombinant protein with a molecular weight of 76000 was expressed. The protein antigen was proved to be tetanus toxin fragment C specific antigen by Western blotting. Animal test showed good immunogenicity of the recombinant antigen. The antigen at a dosage of 1 μg could induce 1 2 IU/ml of antitoxin with an ELISA titer of 1∶800.

  • Clone and Expression of Anti-CD3 Single Chain Antibody Gene

    The genes at the variable regions of heavy (V H) and light (V K) chains of anti CD 3 antibody were linked by splicing, overlapping and extension(SOE). The single chain antibody (ScFv) gene constructed in this way was cloned into the plasmid containing Tac promotor, then recon was selected out by dot blot hybridization and transformed to E. coli BL 21 (DE3). The result showed that anti CD 3 single chain antibody protein was successfully expressed.

  • Neutralization Effect of IgY on Cytotoxic Activity of Helicobacterpylory Somatic Antigen

    The Vac A + helicobacterpylory(HP) strain isolated from patients was lysated by sonication and ultracentrifugation, and the cytotoxic activity of HP somatic protein, as well as the protective effect of the IgY extracted from the egg yolk laid by the hens immunized with the protein, were detected by the in vitro culture of Hela cells. SDS PAGE proved that HP somatic antigen consisted of at least 10 components, and the molecular weight of main components of it ranged from 67 000 to 94 000. The ELISA using HP somatic antigen as coated antigen proved that the serum IgG titers of the patients with duodenal ulcer were significantly improved. The HP somatic protein showed significant cytotoxic activity to Hela cell, with LD 50 less than 12 μg/ml. The activity could be concentration dependently blocked by the IgY extracted from egg yolk laid by the hens immunized with Hp antigen. It indicated that local administration of IgY might be an effective treatment for HP infectious disease.

  • Pharmacological Experiments of Fetal Dog Brain Injection

    The influence of fetal dog brain injection prepared by the authors on brain function was studied by hypoxia tolerance, cerebral ischemia and swimming maze tests in mice and detecting for the malondialdehyde(MDA) content and superoxide dismutase(SOD) activity in murine brain tissue. The result showed that the injection could significantly improve the tolerance of mice to hypoxia induced by potassium cyanide (p<0.01) , prolong the respiration duration of them after decapitation (p<0.001) and reduce the dysmnesia induced by alcohol (p<0.005). It could also significantly decrease the MDA content in murine brain tissue (p<0.001) and improve the SOD activity in the tissue (p<0.001).

  • Transgenic Mice Susceptible to Poliovirus Serves as An Animal Model for Neurovirulence Test of Live Poliomyelitis Vaccine

    A strong poliovirus strain type Ⅲ(Leon) and 3 lots(WHO/Ⅲ,93/363 and 3J) of live poliomyelitis vaccines type Ⅲ were injected into the spinal cord of the transgenic mice carrying human poliovirus receptor(PVRTg21). Clinical trial and histopathological examination showed strong virulence of Leon strain. Paralysis and death were caused by 2.0LogTCID 50 of the strain in 100% of the mice. The virulence of WHO/Ⅲ vaccine was the weakest. Being injected with 5.5 Log TCID 50 of the vaccine,paralysis and death were caused in 81.7% and 51.7% of the mice respectively. Both the virulences of the other 2 lots were higher than that of reference. All the observation indexes of the vaccines with various virulences were significantly different. It proved that PVR Tg21 mice could be used as the animal model for evaluating the virulence test of live poliomyelitis vaccine. The animal could also be applied for virological and epidemiological studies.

  • Selection and Application of McAbs to the Common Epitopes of Mycoplasma

    The authors established 7 hybridoma cell strains using Mhy and Mp as immunogens, and found that 2 of the 7 strains, named AH5 and DD3, could secret McAbs which had strong cross reaction with M.hyorhinis (Mhy), M.arginini(Ma), M.orale(Mo), M.pnenmonia(Mp) and A.Laidlawii(Al). It indicatied that the 2 kinds of antibodies were specific to the common epitopes of the 5 kinds of mycoplasma. A indirect fluoresence method was developed using the antibody secreted by DD3 as the 1st antibody and used for detecting the mycoplasma in Vero cells. The sensitivity of the method(200/ml) was significantly higher than that of culture method and comparable with that of DNA staining method. The mycoplasma in culture supernatant was detected by sandwich ELISA using the antibodies secreted by AH5 and DD3 as coated and enzyme labelled antibodies respectively. The sensitivity of the method was 12.5 ng/ml. Twenty mycoplasma positive and 10 negative cell strains were detected by the 2 methods. The result showed that, by indirect fluoresence method, all the 20 positive and 1 of the 10 negative strains were positive; however, by sandwich ELISA, 18 of the 20 postive strain were positive, and all of the negative strains were negative.\;

  • Preparation and Application of McAbs to pTrcHis Vector Protein

    Two hybridoma cell strains (1C4 And 3C10) secreting McAbs were obtained by immunizing BALB/c mice with the recombinant proteins (rp150c and rp52c) expressed in E.coli using pTrcHis vector. The McAbs secreted by strains 1C4 and 3C10 were IgG 3 and IgM respectively, and the ELISA titers of them in murine ascites were 1∶2×10 6 and 1∶600 respectively. The McAb secreted by strain 1C4 showed good specificity, and no non specific reaction was observed between it and host bacterial protein. A kit was prepared with 1C4 McAb after being labelled with HRP and used for detecting HCMV IgM positive serum. The result showed the the McAb was available.\;

  • Establishment of A New Method for Extracting Streptococcus Protein G

    A new method for extracting streptococcus protein G(SPG) was reported in this paper. SPG was extracted from the cell wall of strain G148 of streptococcus, digested with papain ,then purified by ion exchange and affinity chromatographies. The activity of the SPG was detected by ELISA, and the purity and molecular weight of it were detected by SDS PAGE.

  • Establishment of A Method for Detecting In Vitro Biological Activity of rHu TPO

    A method for detecting the in vitro biological activity of rHu TPO with TPO dependent cell strains was developed. The standards and samples of TPO were diluted to certain concentrations and added to a 96 well tissue culture plate. After TPO dependent cells were added in each well, the plate was cultured in CO 2 incubator at 37℃ for 48 h, and further cultured for 6±2 h after MTS was added in. The in vitro biological activities of samples were calculated out with A 492 values by dose respones paralled line method, using TPO standard as control. The method showed good reproducibility and high recovery rate, and was suitable for various laboratories.

  • Detection of K-ras Gene Mutant in Pancreatic Juice by Nested PCR-RFLP

    In order to identify pancreatic cancer cases,the mutant at the 12th codon of K ras gene in the pancreatic juice from the doubtful patients with pancreatitis and pancreatic cancer were detected by nested PCR RFLP.No K ras gene mutant was detected out in the 11 specimens from the patients with pancreatitis,however,it was detected out in 9(81.2%) of the 11 specimens from those with pancreatic cancer.

  • Comparison of Qualities of Live Attenuated Hepatitis A Vaccine (Strain L-A -1) Prepared by Different Processes

    The qualities of live attenuated hepatitis A(HA) vaccine (strain L A 1) prepared by 2 different processes were compared. In the lst process, the cells were infected with HA at the same time of passage; however, in the 2nd process, they were infected after monolayer was formed. The result showed that, by the 1st process, the production cycle of vaccine was significantly shortened, the virus titer of it was significantly improved, and, even at the peak time of virus proliferation, no significant cytomrphological change was observed. It indicated that the 1st process was more suitable for the large scale production.

  • Curative Effect of Large Dose of Intravenous Ig on Autoimmune Hemolytic Anemia

  • Double-blind Field Trial of Oral Live F2a-Sonne(FS) Dysentery Vaccine

    In order to observe the safety and protective effect of oral live F 2a Sonne(FS) dysentery vaccine, 26230 subjects were randomly divided into 3 groups. The 2 trial groups received high and low doses of the vaccine respectively, and the control group received placebo. The result showed that the FS vaccine was safe and reliable. During the following 6 months after immunization, the protective rates of the FS vaccine against F 2a , Sonne and F 2a +Sonne dysentery were 61.07%, 72.48% and 62.76% respectively, and that against all kinds of bacillary dysentery was 52.02%. However, no significant difference was observed between the protective rates of high and low dose groups.
