  • Study on the Sero-Epidemiologic Efficacy of Shanghai 191 Measles Live Vaccine after 16 Years since Primary Immunization

    This article reports the study of the duration of immunity induced by Shanghai 191 measles live vaccine in Gao Tang Island of Xiangshan County. 273 children immunized in 1973 were observed over 16 consecutive years. The positive rate of measles HI antibody was 83.2% and GMT was 1:3.6 in the 16th year.In an outbreak of natural measles in Gao Tang Island in 1988,34 vaccinees were exposed by patients.Two children who had been vaccinated in 1973 End 1978 became ill with measles (their HI antibody<l:2 in 1987), but 32 children (HI antibody<l:2~1:8 in 1987) were inapparent infection. The results suggested that this vaccine has adequately iminunogenic.

  • The Copy Number of Expression Plasmid in Engineered Strain for Producing HulFNα1

    Under different growth conditions, the copy number of expression plasmid in IFN伪_1 producer BMH71-18/pBV867 was determined by fluorescence technique. The results showed that the copy number of plasmid in IFN伪_1 Producing strain was stable at about 100 copies/cell during high density fermantation. It is about 1.6 fold of that in the cell, which were grown in the shaking flask.The relationship between cell density and expression level in the course of fermantation was discussed.

  • Study of the Immunization Procedures in Young Children with Polysaccharide Vaccine of Meningoccus Groups A

    The serological effect of polysaccharide vaccine of meningoccus groups A with different basic immunization procedures, different interval and differnt dosse waes evaluated in 1,600 children aged from 6 months to 2 years old. The results showed that th immunization program was appropriate with 3 months of the time interval between two njections for basic immunization with 50μg dose and the boosting injection one year ater.

  • Preparation, Physiochemical Characteristics and Stability of Meningococcal Group A Polysaccharide-Tetanus Toxoid Conjugate Antigen

    According to quality requirement of biologics, we prepared three successive lots of Meningococcal Group A Polysaccharide-Tetanus Toxcid Conjugate Vaccine, which was composed of 19.8-24.4% (w/w) Polysaccharide moiety. The neoglycoprotein had a characteristic ultraviolet spectrum of 245nm and Kd value of 0 on Sepharose-CL4B.The immunogenicity of Ps moiety of the conjugate was markedly enhanced although the antigenicity of Ps moiety was the same as plain polysaccharide on the rocket immunoelectrophoresis.The stability of the conjugates was examined. There was no significant difference in immunogenicity of the conjugates during 3-year storage at 4℃ or 8-week storage at 37℃. The tests using gel permeation chromatography with Sepharose-CL4B and SDS-PAGE strongly affirmed the covalency of the derivatized protein-polysaccharide linkage and its stability through high temperature treatment.

  • Adaptation and passages of rabies virus strains in vero cell cultures

    Rabies is a lethal widespread epidemic. At present,only means of prevention and cure of rabies is injection of rabies vaccine.WHO has recently recommended the Purified Vero Rabies Vaccine(PVRV). Vero cells were chosen as a substrate for rabies vaccine production because of the cell growth potential in microcarrier culture and the high virus yield,i.e, conditions suitable for an economical large-scale production of inactivated rabies vaccine.Potency and stability of PVRV have been satisfactory, equivalent to or higher than human diploid cell vaccine.The study and preparation of PVRV have just begun in our country.The important factor in the production of vaccine is to increase the titer of virus, which was studied and explored in this paper.The culture condition and passage methods of CTN-1-42, CTN-1-103, 4aG three rabies virus strains were studied in vero cells. The titer of virus suspension in each passage were determined by mice(i. e)test and plaque formation test after 8 or 9 passages. The results showed that:(l)The mean titer of C42-Vl7 strain was about 7.13 LogLD 50/ml through 17 passages in vero cells, which reached the standard of unnecessary concentration of virus harvest bv WHO regulation. (2) The incubation period in vero cells was shortened from about 10 days to 3~5 days. The titer of other two virus strains(≥10~6.5LogLD_(50)/ml)reachcd the standard for the preparation of PVRV(≥10~6.0Log LD_(50)/ml).

  • Oral Rabies Vaccine for Use in Dogs

    Dogs were immunized orally with an attenuated rabies vaccine, No.22 strain on mice brain.The liquid vaccine was placed in rice porridge,which is acceptable to and readily ingested by the animals.Serological response was tested with mice serum neutralization test.After immunization seroconversion rates in seronegative dogs were 100% (GMT 42, N=20), 91.7%(GMT76, N=12)and 80% (GMT34,N=5) in groups receiving oral vaccine after 1,6 and 15 months respectively.These results indicate that the attenuated oral rabies vaccine virus was highly immunogenic,genetically stable,and safe for dogs.According to the data mentioned above,it is considered that the No. 22 virus strain could be used as a seed virus for ths production of live attenuated rabies vaccine for dogs.

  • Comparison of Various Potency test Methods on Adsorbed purified Tetanus Toxoid

    This article presents the results of standardization of adsorbed purified tetanus toxoid by 3 potency test methods.The method recommended by WHO is admirable in accuracy, but it consumes much laboratory animals. There is no requirements in Chinese Regulation to set up a control group with a corresponding standard vaccine. Thus when the experimental group proceeds alone without a control, the results are usually affected by seasonal and animal feeding conditions. For the time being, the standardization of tetanus toxoid by a method easy to perform and comparatively rational is to immunize animals with the product to be identified and the referencing vaccine simultaneously, and then the titres of antitoxin are tested. If the antitoxic titre in the experimental serum equals or exceeds to that in the control group then the product is up to standard.

  • Detection of MHV Antibody in Experimental Mice and Studies on Its Vertical Transmission

    To examine the quality of mice in open breeding.The mice obtained from several health institutes in cur country were detected for MHV antibody by ELISA from 1982 to 1988,using the SPF mice as control animals.The result proved that the vertical transmission of mouse liver virus might be produced by means of experimental infection or natural infection.

  • Pathological Diagnosis of Mouse-pox and Cellular Localization of the Ectromelia Viral Antigen by Immuno-enzyme Technique

    We have experimentally observed the clinical manifestation and pathological changes caused by natural and experimental mouse-pox infections in mice, described those morphological characteristics of the inclusion body at the skin lesions, liver and some other organs,also suggested the differential diagnosis of mouse-pox from hepatitis, TYZZER's and disease and typhoid in mice. The cellular localization of Ectromelia viral antigen by immunohistochemical technique(PAP)was also studied.
